siRNA-induced DNA methylation in tumorigenesis




siRNA-induced gene silencing is considered the first only in the cytoplasm of the transcriptional regulation of the process, with the siRNA guide the discovery of DNA methylation, has been confirmed through the guide genome siRNA modification caused the apparent transcriptional gene silencing. DNA methylation has been predicted is a carcinogenicity epigenetic mechanism of tumorigenesis involving tumor suppressor gene silencing abnormal gene promoter region DNA methylation. analysis of these two processes inherent relations, and explore siRNA on tumor cells and gene expression of the role. This will help to tumor biology and epigenetics research, also for the development of prevention and treatment of tumors new methods and ways to offer new ideas.

English Abstract:

The most familiar action of mode performed by RNA regulation is the posttra criptional gene silencing in the cytoplasm. The discovery of siRNA-directed DNA methylation in nucleus has proved that siRNA can induce tra criptional gene silencing by siRNA-guided genome modificatio. DNA methylation was once predicted to be an epigenetic mechanism of tumor biogenesis. It is well known that the a ormal inhibition of tumor-su re or genes is a ociated with DNA de novo methylation in the gene promoter region. Potential molecular mechanisms of tumour-su re or gene silencing was analized, the principle of siRNA-directed tra criptional su re ion was explored, and the po ible relatio hip between them was clarified. The detailed study on the relatio hip between DNA methylation and RNA interference will be beneficial to better understand tumor biology and epigenetics, and also provide new therapeutic strategies for the prevention and treatment of cancer.
